Activity Assay

Proteinase K activity can be measured using hemoglobin as the substrate.

Description

Proteinase K belongs to the serine protease family (Peptidase S8) and exhibits broad substrate specificity. Unlike many proteases, Proteinase K efficiently digests proteins in their native folded conformation, making it highly effective for nucleic acid purification and protein removal.

Proteinase K activity can be inhibited by serine protease inhibitors such as PMSF. The enzyme remains stable in buffers containing EDTA and is generally unaffected by sulfhydryl-modifying reagents.

Suggested Working Concentration

Recommended working concentration: 0.05–1 mg/mL depending on sample type and application.

Reconstitution

StemImmune Ready-to-Use Proteinase K does not require reconstitution and can be used directly from the bottle.

Stability & Storage

Store at 15–25°C. Avoid contamination during use. Refer to the Certificate of Analysis for lot-specific information.

Note: For dilution, use nuclease-free water. DEPC-treated water may be used when appropriate for RNA-related applications.
