Components

Note: pH 8.3 ± 0.1 @ 25℃ when prepared as 1× solution.

Description

TAE appears as white instant granules. Each pouch makes 1 L of 1× TAE solution conveniently. The main components are Tris-acetate and EDTA-2Na. The final concentration of 1× TAE is 40 mM Tris-acetate and 1 mM EDTA-2Na.

• Widely used as nucleic acid electrophoresis buffer in biology labs.
• Recommended for agarose gel electrophoresis of DNA, especially for fragments >13 kb.
• Suitable for recovering DNA from gel after electrophoresis.
• Not ideal for long-term electrophoresis runs due to relatively low buffering capacity.

Method

1. Add ~600 ml distilled water and a magnetic stir bar into a beaker.
2. While stirring, slowly add contents of 1 pouch of TAE until dissolved.
3. Add distilled water to bring final volume to 1 L → 1× solution ready to use.
